Prediction of Passive Membrane Permeability of Linagliptin

نویسندگان

چکیده

Linagliptin is a highly selective inhibitor of dipeptidyl peptidase-4 and therefore clinically utilized to treat adults with type 2 diabetes mellitus. The objective this study was predict how the ions normally found in extracellular or intracellular fluids within body system will influence linagliptin’s rate passive diffusion across cell membrane. methodology involved measuring linagliptin partition coefficient chloroform-water containing salts at 25 oC by shake flask method. results indicate that highest concentration (0.5 M) studied, sodium chloride give value when compared control. In conclusion, physiological would have little no effect on drug’s molecular state fluid as failed significantly alter drug, hence not membrane permeability linagliptin.

Modeling the pharmacodynamics of passive membrane permeability

Small molecule permeability through cellular membranes is critical to a better understanding of pharmacodynamics and the drug discovery endeavor. Such permeability may be estimated as a function of the free energy change of barrier crossing by invoking the barrier domain model, which posits that permeation is limited by passage through a single "barrier domain" and assumes diffusivity differenc...

Passive ion permeability of the chromaffin-granule membrane.

'Ghosts' of bovine chromaffin granules, in which the complex mixture of proteins and solutes normally found in the granule matrix is replaced by buffered sucrose are osmotically sensitive. They shrink when the osmotic pressure of the suspension medium is increased, and swell if solute entry is facilitated by the addition of ionophores. Swelling in the presence of ionophores has been used to inv...

pH-SPECIFIC PREDICTION MODELS FOR MEMBRANE PERMEABILITY

Approved drugs include about 85% of ionisable compounds. Majority of drug-related properties in the early stage of drug research are measured at the neutral pH, while properties of ionisable compounds depend on the pH of the environment. The pH range in gastrointestinal tract varies in the large scale (from pH ~2 to ~9) and therefore influences human intestinal absorption. The robust experiment...

Prediction of the Passive Intestinal Absorption of Medicinal Plant Extract Constituents with the Parallel Artificial Membrane Permeability Assay (PAMPA).

At the early drug discovery stage, the high-throughput parallel artificial membrane permeability assay is one of the most frequently used in vitro models to predict transcellular passive absorption. While thousands of new chemical entities have been screened with the parallel artificial membrane permeability assay, in general, permeation properties of natural products have been scarcely evaluat...
